The bagrid catfish, Pseudobagrus ussuriensis, exhibits significant sexual dimorphism in growth rate and body size with males growing faster than females. Therefore, an all‐male culture can dramatically increase the output and profitability of fishery products. According to the monosex breeding route, super‐male individuals’ acquirement by XY male sex reversal and artificial gynogenesis is the key step. An effective protocol to induce meiotic gynogenesis using homologous sperms has been developed in this study. The most effective UV irradiation for sperm genetic inactivation was found to be at a distance of 20 cm with 66 μW/cm2 light intensity for 25 min. Optimal treatment for cold shock was 5 min post‐fertilization at 0‐4°C for 30 min, which gave the best survival rate of 13.65 ± 2.87%. The sex ratio in the meiotic gynogens showed a significant female‐biased deviation (p < .05); thirty meiogynogens and their parents were further analysed using a male‐specific AFLP marker, of which only the male parent produced a male‐specific DNA band of 412 bp. These results indicated the female homogametic XX/XY sex determination system in P. ussuriensis. The optimization of a protocol for the successful induction of meiogynogenesis in the bagrid catfish lays the basis for all‐male production and is useful in ascertaining the genetic sex determination system in this promising aquaculture species. 相似文献

The stabilities of seven agronomic traits were analyzed and the general stabilities of soybean [Glycine max (L.) Merr.] accessions were evaluated based on the additive main effects and multiplicative interactions (AMMI) model using the founder parent Tokachi nagaha and 137 of its derived cultivars as materials. The objective was to provide a theoretical basis for effectively using germplasm in soybean breeding and production. Analysis of variance (ANOVA) showed that genotype, environment, and genotype by environment interactions were significantly different for each trait. The first three interaction principal components axes (IPCA) were highly significant, accounting for 61.28–70.00% of the total variation. The stability differed for the different traits. 50 cultivars with high general stabilities were identified. The general stability of Tokachi nagaha was moderate, as the stability coefficients (Di) of its seven traits were relatively high; this must be considered by breeders using this cultivar as a breeding parent. There were significant positive correlations between the phenotypic values and their own Di values for number of branches per plant, number of pods per plant, number of seeds per plant, and seed weight per plant. This would lead to the expectation that the phenotypic stability would be lower when a cultivar had more branches, pods, seeds, and high per plant yields. Background
Male fertility is crucial for rice yield, and the improvement of rice yield requires hybrid production that depends on male sterile lines. Although recent studies have revealed several important genes in male reproductive development, our understanding of the mechanisms of rice pollen development remains unclear.Results
We identified a rice mutant oslap6 with complete male sterile phenotype caused by defects in pollen exine formation. By using the MutMap method, we found that a single nucleotide polymorphism (SNP) variation located in the second exon of OsLAP6/OsPKS1 was responsible for the mutant phenotype. OsLAP6/OsPKS1 is an orthologous gene of Arabidopsis PKSA/LAP6, which functions in sporopollenin metabolism. Several other loss-of-function mutants of OsLAP6/OsPKS1 generated by the CRISPR/Cas9 genomic editing tool also exhibited the same phenotype of male sterility. Our cellular analysis suggested that OsLAP6/OsPKS1 might regulate pollen exine formation by affecting bacula elongation. Expression examination indicated that OsLAP6/OsPKS1 is specifically expressed in tapetum, and its product is localized to the endoplasmic reticulum (ER). Protein sequence analysis indicated that OsLAP6/OsPKS1 is conserved in land plants.Conclusions
OsLAP6/OsPKS1 is a critical molecular switch for rice male fertility by participating in a conserved sporopollenin precursor biosynthetic pathway in land plants. Manipulation of OsLAP6/OsPKS1 has potential for application in hybrid rice breeding.54.
为研究不同品种藜麦幼苗对海拔梯度变化的生理响应机制,本试验以4个藜麦品种—‘陇藜1号’、‘陇藜2号’、‘陇藜3号’及‘陇藜4号’为材料,选取甘南州临潭县3个不同海拔(2380 m,2580 m,2780 m)分布区为试验地。通过测定幼苗生物量及生理生化指标,分析不同藜麦品种幼苗在不同海拔试验区的生理响应机制,为建立藜麦品种和环境相适宜的评价体系提供参考依据。结果表明,随海拔升高,藜麦叶片叶绿素、可溶性蛋白以及抗坏血酸过氧化物酶(APX)含量逐渐降低,海拔2780 m最低;超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD)逐渐升高,在海拔2780 m达到最大值;超氧阴离子(O2·-)产生速率呈现先升高后降低的趋势。不同藜麦品种幼苗生物量随着海拔的升高逐渐降低。说明高海拔下太阳辐射强度增大、温度降低,导致藜麦幼苗植株抗氧化酶活性升高以清除多余活性氧(ROS)自由基,使得植株能够适应高海拔环境而正常生长,但生物量较低海拔仍有降低。因此,不同品种藜麦在低海拔地区受到的胁迫较低,虽然在高海拔区域胁迫加剧,但藜麦幼苗可以正常生长并进行干物质的积累。 相似文献

以滇东北4种典型筇竹林为研究对象,对比分析不同类型筇竹林凋落物储量及持水性能,从而为滇东北筇竹林生态可持续经营提供理论依据和实践指导。研究结果表明,凋落物储量为人工筇竹-黄皮树混交林(7.61 t/hm2) > 天然筇竹-人工黄皮树混交林(6.61 t/hm2) > 天然筇竹-人工厚朴混交林(5.73 t/hm2) > 天然筇竹纯林(5.23 t/hm2)。凋落物最大持水量为天然筇竹-人工厚朴混交林(3.45 t/hm2) > 人工筇竹-黄皮树混交林(3.22 t/hm2) > 天然筇竹-人工黄皮树混交林(2.89 t/hm2) > 天然筇竹纯林(2.69 t/hm2)。4种类型筇竹林凋落物吸水速率均随着浸泡时间延长而逐渐趋于一致,筇竹混交林凋落物吸水速率高于筇竹纯林。凋落物的总有效拦蓄量为人工筇竹-黄皮树混交林(9.74 t/hm2) > 天然筇竹-人工厚朴混交林(8.95 t/hm2) > 天然筇竹-人工黄皮树混交林(7.73 t/hm2) > 天然筇竹纯林(6.23 t/hm2)。 相似文献

为探究播期对直播水稻产量、花后干物质和氮素积累与转运的影响,以水稻品种白粳1号、长白9号和龙粳31号为供试材料,设置3个播期(SD1、SD2和SD3),比较不同播期条件下3个品种的产量、抽穗期和成熟期各器官干物质及氮素积累与转运特点。3个品种抽穗期和成熟期各器官干物质及氮素积累量、抽穗至成熟期各器官干物质及氮素转运量和产量均表现为SD2>SD3>SD1。相关分析表明,产量与成熟期穗干物质及氮素积累量呈显著正相关,与叶片干物质和氮素转运量呈显著正相关。在本试验条件下,播期的推迟提高了3个品种有效穗数及结实率,进而提高了产量,同时促进了各器官物质转运量与转运率的提高。各品种SD2处理的产量及花后干物质、氮素积累量最高,SD2为适宜播期。 相似文献
